Resumen : | El cultivo del nogal (Juglans regia L.), es el segundo frutal más cultivado en Chile,
concentrándose alrededor del 25 % de superficie plantada entre las regiones del
Maule y la Araucanía, donde este frutal es afectado severamente por la enfermedad
Peste negra, causada por la bacteria Xanthomonas arboricola pv. juglandis (Xaj).
En el control de esta enfermedad se utilizan antibióticos, los cuales pueden generar
resistencia en las cepas bacterianas. En esta investigación se evaluó la diversidad
genética presente en 124 aislados bacterianos provenientes de yemas de seis
huertos ubicados entre las regiones de Ñuble y Biobío, mediante el estudio de la
huella genética molecular usando BOX PCR. Se observó una gran variabilidad
genética de los aislados bacterianos de Xaj obtenidos desde los seis huertos
evaluados, identificando la presencia y ausencia de bandas de tamaños
moleculares específicos. Además, se evaluó in vitro la resistencia de los aislados a
distintos antibióticos utilizados para el control de esta enfermedad a base de:
Clorhidrato de Tetraciclina, Sulfato de Estreptomicina y Clorhidrato de
Kasugamicina. El 78 % de las cepas de Xaj presentaron resistencia a algún
antibiótico, en donde el mayor número de cepas resistentes se presentó al utilizar
Clorhidrato de Kasugamicina, siendo menor al utilizar Sulfato de Estreptomicina y
Clorhidrato de Tetraciclina o su mezcla. Walnut (Juglans regia L.) is the second most cultivated fruit tree in Chile, concentrating around 25 % of the planted area between the Maule and Araucanía regions, where this fruit tree is severely affected by the Black Plague disease, caused by the bacterium Xanthomonas arboricola pv. juglandis (Xaj). Antibiotics are used to control this disease, which can generate resistance in bacterial strains. In this research, the genetic diversity of 124 bacterial isolates from buds of six orchards located between the Ñuble and Biobío regions was evaluated through molecular fingerprinting using BOX PCR. A great genetic variability of the Xaj bacterial isolates obtained from the six evaluated orchards was observed. In addition, the antibiotic resistance of the strains to different antibiotics used for the control of this disease was assessed in vitro. The antibiotics were Tetracycline Hydrochloride, treptomycin Sulfate and Kasugamycin Hydrochloride. 78 % of the Xaj strains showed resistance to some antibiotic, where the highest number of strains were resistant to Kasugamycin Hydrochloride, and the lowest number of resistant strains where observed when using Streptomycin Sulfate and Tetracycline Hydrochloride or their mixture. |
