El microambiente tumoral del OSCC induce una alteración del linfocito T, promoviendo un fenotipo exhausto y una reprogramación metabólica. El organelo principal del metabolismo es la mitocondria y en los últimos años se ha indicado que diversas células tienen la capacidad de transferir mitocondrias, incluyendo la cancerosa. Sin embargo, a la fecha no se ha evaluado si existe la transferencia de mitocondrias desde células cancerosas hacia los linfocitos T y si tal fenómeno explica el fenotipo exhausto del linfocito T. El objetivo de este trabajo fue analizar el fenotipo exhausto en linfocitos TCD4+ post transferencia artificial de mitocondrias (MitoCeption) obtenidas desde la línea celular de cáncer oral HSC-3. La metodología se basó en la estandarización de la transferencia mitocondrial artificial utilizando mitocondrias aisladas marcadas con MitoTracker desde células tumorales de cáncer oral sobre linfocitos T CD4+ a través de MitoCeption. Luego, se analizó la expresión de moléculas de superficie, proliferación y secreción de citoquinas mediada por transferencia mitocondrial tumoral a través de citometría de flujo. En los resultados se observó que los linfocitos TCD4+ que adquirieron mitocondrias presentaron un alza en la expresión de 2 proteínas inhibidoras (TIGIT y CTLA4) y 3 asociadas con fenotipo exhausto (PD-1, PLD-1 y LAG3), en comparación con el grupo control (p: 0,0184; p: 0,0299; p: 0,0088; p: 0,0205; p: 0,0232 respectivamente). Además, los linfocitos mitoceptados exhibieron una disminución significativa en la proliferación en comparación con las células control, no mitoceptadas. Para el análisis de citoquinas, se observó una disminución significativa en el grupo mitoceptado en la secreción de IFN-gamma, TNF-alpha, IL-10 e IL-4, citoquinas de la vía TH1 y TH2 (p: 0,0149; p: 0,0149; 0,0150; p: 0,0353 respectivamente) en comparación con el control. Por lo tanto, se concluyó que la adquisición de mitocondrias aisladas desde células cancerosas HSC-3 por el linfocito T CD4+ induce efectos a nivel funcional induciendo un alza en los receptores inhibitorios y de fenotipo exhausto e inhibiendo la proliferación y la secreción de citoquinas tipo TH1 y TH2. Estas características son de un fenotipo exhausto, por ende, se concluye que la transferencia de mitocondrias desde una célula cancerosa induce un fenotipo exhausto en el linfocito TCD4+.Oral squamous cell carcinoma (OSCC) is the most frequent type of oral cancer in Chile and in the world. The tumor microenvironment of OSCC induces an alteration of the T lymphocyte, promoting an exhausted phenotype and metabolic reprogramming. The main organelle of metabolism is the mitochondrion and in recent years it has been indicated that several cells have the capacity to transfer mitochondria, including cancer cells. However, to date, it has not been evaluated whether mitochondria transfer from cancer cells to T lymphocytes exists and whether such a phenomenon explains the exhausted phenotype of the T lymphocyte. The aim of this work was to analyze the exhausted phenotype in TCD4+ lymphocytes after artificial transfer of mitochondria (MitoCeption) obtained from the oral cancer cell line HSC-3. The methodology was based on the standardization of artificial mitochondrial transfer using isolated MitoTracker labeled mitochondria from oral cancer tumor cells onto CD4+ T lymphocytes via MitoCeption. Then, surface molecule expression, proliferation and cytokine secretion mediated by tumor mitochondrial transfer were analyzed by flow cytometry. The results showed that TCD4+ lymphocytes that acquired mitochondria had increased expression of 2 inhibitory proteins (TIGIT and CTLA4) and 3 proteins associated with exhausted phenotype (PD-1, PLD-1 and LAG3), compared to the control group (p: 0.0184; p: 0.0299; p: 0.0088; p: 0.0205; p:0.0232, respectively). In addition, the mitocepted lymphocytes exhibited a significant decrease in proliferation compared to the control, non-mitocepted cells. For cytokine analysis, a significant decrease was observed in the mitocepted group in the secretion of IFN-gamma, TNF-alpha, IL-10 and IL-4, TH1 and TH2 pathway cytokines (p: 0.0149; p: 0.0149; 0.0150; p: 0.0353 respectively) compared to control. Therefore, it was concluded that the acquisition of isolated mitochondria from HSC-3 cancer cells by CD4+ T lymphocyte induces effects at the functional level by inducing an upregulation of inhibitory and exhausted phenotype receptors and inhibiting proliferation and secretion of TH1- and TH2 type cytokines.
