The progressive increases in the demand of the communities, the market and the challenges posed by climate change demand greater efforts in the search for effectiveness and efficiency in the cultivation processes oriented to the production of wood. Given the multiple limiting factors for genetic improvement, coming from the ignorance of the molecular processes involved in the development of Eucalyptus, it is necessary to propose new methodologies to reveal the molecular machinery involved in the abiotic stress response. The present study sought to reveal the influence of long-chain non-coding RNAs (lncRNAs) on the response to lowtemperature stress. The proposed methodology sought to complement the strengths and weaknesses of different bioinformatics tools to elucidate the role of lncRNAs in the response to low-temperature stress in a Eucalyptus globulus x Eucalyptus nitens hybrid. As a result, 22 lncRNAs were identified that could participate in the modulation of the cold responsive genes associated in the hybrid. Based on the results and the information associated with analyzes in other plant species, it is concluded that, based on the abundance of lncRNA transcripts and the in silico expression of the putative target genes, there would be regulation of gene expression in response to stress by low temperatures by the lncRNAs.
