Análisis de los residuos relevantes en la coordinación con el cofactor Manganeso en la Agmatinase Like protein (ALP).

dc.description.abstractLa agmatinase like protein (ALP) es una enzima que se clonó a partir de una librería de genes de cerebro de rata y presenta actividad agmatinasa, hidrolizando la agmatina a putrescina y urea. Esta enzima requiere Mn2+ como cofactor y posee un bajo porcentaje de identidad con las agmatinasas conocidas, no conservando los residuos de coordinación al metal activador. En trabajos previos del laboratorio por la falta de información estructural se realizó un modelo estructural de ALP, en el cual se propusieron 5 posibles residuos que interaccionarían con el cofactor Mn2+. Se generó una mutante triple (N213A/Q215A/D217A) y una mutante doble (E288A/K290A) de estos residuos y ambas mutantes no presentaron actividad enzimática. Esta tesis tiene como objetivo evaluar la participación de cada uno de los residuos de la mutante triple en la actividad y coordinación metálica de ALP. Para ello, se generaron mutantes simples reemplazando cada residuo por alanina a través de mutagénesis sitio dirigida, se purificó parcialmente cada enzima por cromatografía de intercambio aniónico y se analizaron sus parámetros cinéticos. Como resultado, se obtuvo que las tres mutantes simples presentaron valores de Km y Vmáx menores que la enzima silvestre. Para analizar la interacción enzima-Mn2+, se realizaron experimentos de activación enzimática, diálisis, hiperactivación, determinación del contenido metálico y especificidad de la activación con el metal y en todos los resultados las tres mutantes simples no mostraron diferencias con la enzima silvestre. Concluimos que las tres mutaciones simples afectaron el sitio activo de la enzima ya que alteraron sus parámetros cinéticos y postulamos que estos residuos se encuentran próximos al sitio activo. Dado que las mutaciones no alteraron la interacción enzima-Mn2+ concluimos que estos residuos no participan directamente como ligandos coordinantes del metal activador Mn2+.es
dc.description.abstractThe agmatinase-like protein (ALP) is an enzyme cloned from a rat brain gene library that exhibits agmatinase activity, hydrolyzing agmatine to putrescine and urea. This enzyme requires Mn2+ as a cofactor and has a low percentage of identity with known agmatinases, not conserving the coordination residues to the activating metal. In previous laboratory work, due to the lack of structural information, a structural model of ALP was constructed, proposing 5 possible residues that would interact with the Mn2+ cofactor. A triple mutant (N213A/Q215A/D217A) and a double mutant (E288A/K290A) of these residues were generated, both of which showed no enzymatic activity. This thesis aims to evaluate the participation of each residue in the triple mutant in the activity and metal coordination of ALP. For this purpose, single mutants were generated by replacing each residue with alanine through site-directed mutagenesis, each enzyme was partially purified by anion exchange chromatography, and their kinetic parameters were analyzed. As a result, all three single mutants showed lower Km and Vmax values compared to the wild type enzyme. To analyze the enzyme-Mn2+ interaction, experiments including enzymatic activation, dialysis, hyperactivation, determination of metal content, and specificity of activation with the metal were conducted, and in all cases, the three single mutants showed no differences compared to the wild-type enzyme. We concluded that the three single mutations affected the enzyme's active site as they altered its kinetic parameters, and we postulate that these residues are located near the active site. Since the mutations did not alter the enzyme-Mn2+ interaction, we conclude that these residues do not directly participate as coordinating ligands for the activating metal Mn2+.en
